I’m running a set of Michelin Pilot Sport AS3+. They have less than 25K on them. I had to replace one of the rears due to a puncture. Will that cause any problem with the dif being it’s slightly larger?

A new tyre has 8mm
If the old has less than 5mm would have replaced it.

Never ever have different tyres on the same axle. Ideally not different brands but certainly not different sizes/profiles.
